Objetivo: O objetivo deste trabalho é avaliar a expressão da molécula de adesão intercelular-1 (ICAM-1) na esclera e coroide de coelhos hipercolesterolêmicos. Métodos: Coelhos New Zealand foram organizados em dois grupos: GN (grupo dieta normal), composto por 8 coelhos (8 olhos), recebeu ração padrão para coelhos, durante 4 semanas; GH (grupo hipercolesterolêmico), composto por 13 coelhos (13 olhos), recebeu dieta rica em colesterol a 1% por 8 semanas. Foi realizada a dosagem sérica de colesterol total, triglicerídeos, HDL colesterol, glicemia de jejum no início do experimento e no momento da eutanásia. Ao final da 4ª semana para o GN e 8ª semana para o GH foi realizada a eutanásia dos animais. Os olhos foram corados com hematoxilina-eosina e submetidos à análise histológica, histomorfométrica e imunohistoquímica com o anticorpo ICAM-1. Resultados: Observou-se significativo aumento do colesterol total e triglicerídeos do GH em relação ao GN (p<0,001). A avaliação histológica com hematoxilina eosina revelou grande quantidade de macrófagos no complexo esclero-coroidal do GH. No GH constatou-se significativo aumento da espessura da esclera e coroide em relação ao GN (p<0,001). Houve significativo aumento da expressão da ICAM-1 na esclera e coroide dos animais do GH em relação ao GN (p<0,001). Conclusão: Este estudo demonstra que a dieta hipercolesterolêmica induz ao aumento da expressão da ICAM-1 na esclera e coroide de coelhos. .


Objective: The aim of this study is to investigate the expression of the intercellular adhesion molecule 1 (ICAM-1) in the sclera and choroid of hypercholesterolemic rabbits Methods: New Zealand rabbits were divided into two groups: the normal diet group (NG), with 8 rabbits (8 eyes), was fed a standard rabbit diet for 4 weeks; the hypercholesterolemic group (HG), with 13 rabbits (13 eyes), was fed a 1% cholesterol- enriched diet for 8 weeks. Total serum cholesterol, triglyceride, HDL cholesterol and fasting blood glucose exams were performed at the start of the experiment and at the euthanasia time. HG and NG animals were euthanized after 8th week and 4th week, respectively. Their eyes were stained with hematoxylin-eosin and underwent histological, histomorphometric and immunohistochemical analyses with ICAM-1 antibody. Results: The diet has induced a significant increase in total cholesterol and triglyceride levels in HG when compared with NG (p<0.001). The histological analysis with hematoxylin-eosin revealed a large number of macrophages in the HG sclera-choroid complex. Moreover, a significant increase in the HG sclera and choroid thickness was observed in relation to NG (p<0.001). There was a significant increase in the ICAM-1 expression in HG sclera and choroid in relation to NG Conclusion: This study has revealed that the hypercholesterolemic diet induces an increase in the ICAM-1 expression in the rabbits’ sclera and choroid. .

OBJECTIVE: To evaluate the effect of blocking the angiotensin II AT-1 receptor by the systemic administration of candesartan on the expression of intercellular adhesion molecule-1 in the sclera and choroid of hypercholesterolemic rabbits. METHODS: New Zealand rabbits were divided into 3 groups, as follows: GI, which was fed a rabbit standard diet; GII, which was fed a hypercholesterolemic diet; and GIII, which received hypercholesterolemic diet plus candesartan. Samples of the rabbits' sclera and choroid were then studied by hematoxylin-eosin staining and histomorphometric and immunohistochemical analyses for intercellular adhesion molecule-1 expression. RESULTS: Histological analysis of hematoxylin- and eosin-stained sclera and choroid revealed that macrophages were rarely present in GI, and GII had significantly increased macrophage numbers compared to GIII. Moreover, in GII, the sclera and choroid morphometry showed a significant increase in thickness in comparison to GI and GIII. GIII presented a significant increase in thickness in relation to GI. Sclera and choroid immunohistochemical analysis for intercellular adhesion molecule-1 expression revealed a significant increase in immunoreactivity in GII in relation to GI and GIII. GIII showed a significant increase in immunoreactivity in relation to GI. CONCLUSION: Candesartan reduced the expression of intercellular adhesion molecule-1 and consequently macrophage accumulation in the sclera and choroid of hypercholesterolemic rabbits. .

The sclera is the outermost layer of the eye. In some vertebrates, it consists of a scleral ring of osseous plates that afford protection against pressure and help maintain the shape of the eyeball. The morphology, number, development and position of the scleral ossicles differ in distinct groups of vertebrates. Therefore, the objective here was to examine the number, shape and arrangement of the scleral ossicles in Brazilian birds of different orders. The study involved 208 birds of 18 orders, which died from causes unrelated to eye infections. The birds’ eyeballs were removed and subjected to diaphanization and staining of the ossicles with Alizarin red S. All the eyeballs analyzed presented scleral ossicles in a fixed position in the central portion of the eyeball, but which varied in size, shape and number. The number of bony plates in the various bird species varied from 11 to 16, with a modal number of 14, and their shape was predominantly quadrangular, showing a similar pattern in species of the same order. Some specimens presented differences in the number of ossicles in the right and left eyeball, as in Spheniscus magellanicus, Elanus leucurus, Ramphastos toco and others and Leptodon cayanensisalso presented a scleral sesamoid bone. The distribution and morphology of the ossicles vary according to the taxonomic group, although species of the same order present a similar pattern.

Na reunião da Associação Pan-americana de Patologia Oftálmica, realizada em Los Angeles CA, 10 de Outubro de 1991, no DOHENY EYE INSTITUTE, C.O. Degrazia propôs o nome de LOFONEUROGONIOMA para um tumor solitário intra-ocular, indiferenciado, originado nas células da lâmina fusca, com células indiferenciadas de expressiva diferenciação para a linhagem schwannocítica, melanocítica e neuroendócrina. Em face do trimorfismo, o patologista pode ser conduzido para os diagnósticos de melanoma, schwannoma maligno ou outro tipo de tumor. O exaustivo estudo do tumor apresentado, através da microscopia eletrônica, da histoquímica e da imunohistoquímica permitiu a formulação da seguinte hipótese: uma célula indiferenciada em repouso, a lofoneurogô- nia, segue as linhagens melanocítica, schwannocítica e neuroendócrina. A diversidade de células dentro de um tumor, o continuus intratumor, responsável pela estrutura em mosaico de muitas neoplasias, além da multiclonalidade resultante de mitoses atípicas, pode ser explicada por essa hipótese. É dessa maneira que se torna compreensível a classificação de Callender para os melanomas intra-oculares nos tipos celulares fusiforme A, fusiforme B, epitelióide, fasciculado e misto, num verdadeiro continuus intertumores. Para justificar a designação proposta, e para enquadrar o caso num grupo taxonômico, foram usadas duas bases classificatórias: 1o ­ histogênica, isto é, correlacionar as células do tumor com as células normalmente existentes nas membranas oculares, no caso, a lâmina fusca; 2o ­ embriogênica isto é, delimitar um grupo de tumores cuja base, no desenvolvimento do embrião, é a crista neural (AU)


In the meeting of the Panamerican Association of Ophtalmic Pathology, held in the Doheny Eye Institute in Los Angeles, CA on Oct 10 1991, C.O. Degrazia proposed the name LOPHONEUROGONIOMA for an undifferentiated intraocular solitary tumor, originating from the cells of the lamina fusca, with undifferentiated cells of expressive differentiation for the schwannian, melanocitic and neuroendocrine lines. Because of the trimorphism, the pathologist may be led to the diagnosis of melanoma, malignant schwannoma or other type of tumor. The exhaustive investigation of the presented tumor through electronmicroscopy, histochemistry, and immunohistochemistry allowed the formulation of the following hypothesis: an undifferentiated cell at rest, the lophoneurogonia, follows the melanocitic, schwannian, and neuroendocrine lines. The diversity of cells inside a tumor responsible for the mosaic structure of many neoplasias, besides the multiclonality resulting from atypical mitoses, can be explained by this hypothesis. This also elucidates Callender's classification of intraocular melanomas in cell types fusiform A, fusiform B, epithelioid, fasciculated and mixed, in a true intertumor continuum. In order to justify the proposed designation, and to fit the case into a taxonomic group, two classificatory bases were used: first, a histogenic one, correlating tumor cells with normally existing cells; and second, embriogenic which bases is the neural crest (AU)

RESUMO

PURPOSE: To compare the ultrasound biomicroscopic measurement of the anterior chamber angle in Asian Indian eyes, with the angle width estimated by gonioscopy. MATERIALS AND METHODS: Participants: Patients with open and closed angles attending a glaucoma clinic were recruited for the study. OBSERVATION PROCEDURES: Temporal quadrants of the angles of patients were categorized by gonioscopy as Grade 0 to Grade 4, using Shaffer's classification. These angles were quantified by ultrasound biomicroscopy (UBM) using the following biometric characteristics: Angle opening distance at 250 micro (AOD 250) and 500 micro (AOD 500) from the scleral spur and trabecular meshwork-ciliary process distance (TCPD). The angles were further segregated as "narrow angles" (Schaffer's Grade 2 or less) and "open angles" (Schaffer's Grade 3 and 4). MAIN OUTCOME MEASURES: The UBM measurements were computed in each case and analyzed in relation to the gonioscopic angle evaluation. RESULTS: One hundred and sixty three eyes of 163 patients were analyzed. One hundred and six eyes had "narrow angles" and 57 eyes had "open angles" on gonioscopy. There was a significant difference among the mean UBM measurements of each angle grade estimated by gonioscopy (P < 0.001). The Pearson correlation coefficient between all UBM parameters and gonioscopy grades was significant at the 0.01 level. The mean AOD 250, AOD 500 and TCPD in narrow angles were 58+/-49 micro, 102+/-84 micro and 653+/-124 respectively, while it was 176+/-47 micro, 291+/-62 micro and 883+/-94 micro in eyes with open angles (P < 0.001) respectively. CONCLUSIONS: The angle width estimated by gonioscopy correlated significantly with the angle dimensions measured by UBM. Gonioscopy, though a subjective test, is a reliable method for estimation of the angle width.
